Product Overview
Clone 4C12 is a mouse monoclonal antibody that recognizes cell surface glycoprotein MUC18 (gene MCAM) in human samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 71.6 kDa, and the supplier reports an observed band at about 120 kDa.
MCAM (CD146, also called MUC18) is an immunoglobulin superfamily cell adhesion molecule expressed on endothelial cells, smooth muscle cells and some activated T cells. It takes part in cell-cell adhesion, endothelial permeability and angiogenesis. MCAM was first identified as a marker of melanoma progression and is studied in vascular biology, tumor metastasis and inflammation.

Samples with a confirmed band (WB): human A375 cells, human HeLa cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human placenta tissue, human mammary cancer tissue, human rectal cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (IHC): human placenta tissue.
Samples with a confirmed signal (flow cytometry): SiHa cells.
Immunogen
Recombinant human CD146 fragment (His59–Ala401), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 73%.
Reactivity Notes
The supplier lists reactivity with human. UniProt places cell surface glycoprotein MUC18 in the membrane. Tissue expression noted by UniProt (human): Detected in endothelial cells in vascular tissue throughout the body. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
