Biological background

Protein function (datasheet): Plays a role in cell adhesion, and in cohesion of the endothelial monolayer at intercellular junctions in vascular tissue. Its expression may allow melanoma cells to interact with cellular elements of the vascular system, thereby enhancing hematogeneous tumor spread. Could be an adhesion molecule active in neural crest cells during embryonic development. Acts as surface receptor that triggers tyrosine phosphorylation of FYN and PTK2/FAK1, and a transient increase in the intracellular calcium concentration. .

Cellular localization (datasheet): Membrane; Single-pass type I membrane protein.

Tissue details (datasheet): Detected in endothelial cells in vascular tissue throughout the body. May appear at the surface of neural crest cells during their embryonic migration. Appears to be limited to vascular smooth muscle in normal adult tissues. Associated with tumor progression and the development of metastasis in human malignant melanoma. Expressed most strongly on metastatic lesions and advanced primary tumors and is only rarely detected in benign melanocytic nevi and thin primary melanomas with a low probability of metastasis.

Research relevance and current trends

  • Commonly studied in contexts related to Cancer,Cell Adhesion,Cell Adhesion Molecules,Cell Type Markers,Cytoskeleton/ECM,Immunology,Invasion/Microenvironment,Signal Transduction,Stem Cells.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Immunofluorescence / ICC: Visualize subcellular localization and co-localization patterns; consider fixation/permeabilization compatibility and controls.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.

Our team were well pleased with the WB result of your anti-CD146 Rabbit Monoclonal antibody. However we have observed positive staining in popliteal artery membrane using this antibody. Is that expected? Could you tell me where is MCAM supposed to be expressed?
According to literature, popliteal artery does express MCAM. Generally MCAM expresses in membrane. Regarding which tissues have MCAM expression, here are a few articles citing expression in various tissues: Cervix carcinoma, Pubmed ID: 18669648, 20068231 Liver, Pubmed ID: 19159218 Melanoma, Pubmed ID: 2602381, 8378324 PNS, Pubmed ID: 15489334 Trachea, Pubmed ID: 14702039
