Product Overview
This rabbit polyclonal antibody targets scavenger receptor cysteine-rich type 1 protein M130 (gene Cd163) in mouse and rat samples and is validated for Western blot, IHC, flow cytometry and ELISA. The predicted molecular weight is 120.9 kDa, and the supplier reports an observed band at about 150–160 kDa.
CD163 is a scavenger receptor on monocytes and macrophages that takes up hemoglobin–haptoglobin complexes, protecting tissues from free hemoglobin and supporting iron recycling. It is widely used as a marker of anti-inflammatory (M2-like) macrophages, and its soluble form is measured as a readout of macrophage activation.

Samples with a confirmed band (WB): rat spleen tissue, rat PC-12 cells, mouse thymus tissue, mouse RAW264.7 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 8% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat thymus tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with a confirmed signal (flow cytometry): RAW264.7 cells.

Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places scavenger receptor cysteine-rich type 1 protein M130 in the [Soluble CD163]: Secreted and SUBCELLULAR LOCATION: Cell membrane. Tissue expression noted by UniProt (mouse): Expressed in monocytes and mature macrophages such as Kupffer cells in the liver, red pulp macrophages in the spleen and mesenteric lymph nodes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
