Overview

This antibody is intended for detection of ALCAM (CD166 antigen) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-CD166/ALCAM Antibody Picoband® catalog # A01788-1. Tested in Flow Cytometry, IF, IHC, WB applications. This antibody reacts with Human, Mouse, Rat. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E.coli-derived human CD166/ALCAM recombinant protein (Position: N167-E406). Human CD166/ALCAM shares 90.8% amino acid (aa) sequence identity with both mouse and rat CD166/ALCAM. (reported region: N167-E406).
  • Molecular weight context: reported MW: 100-110 kDa; calculated MW: 65102 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: Flow Cytometry, IF, IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

CD166 antigen; CD166 antigen. This gene encodes activated leukocyte cell adhesion molecule (ALCAM), also known as CD166 (cluster of differentiation 166), which is a member of a subfamily of immunoglobulin receptors with five immunoglobulin-like domains (VVC2C2C2) in the extracellular domain. This protein binds to T-cell differentiation antigene CD6, and is implicated in the processes of cell adhesion and migration. Multiple alternatively spliced transcript variants encoding different isoforms have been found. Functional note: Cell adhesion molecule that binds to CD6. Involved in neurite extension by neurons via heterophilic and homophilic interactions. May play a role in the binding of T- and B-cells to activated leukocytes, as well as in interactions between cells of the nervous system. Reported localization: Membrane; Single-pass type I membrane protein. Expression/tissue context: Spleen, placenta, liver, and weakly in liver. Expressed by activated T-cells, B-cells, monocytes and thymic epithelial cells. Expressed by neurons in the brain. Restricted expression in tumor cell lines. Preferentially expressed in highly metastasizing melanoma cell lines. .

Research relevance and current trends

  • Adaptive Immunity: Researchers commonly examine how ALCAM (CD166 antigen) relates to this theme using model systems and orthogonal readouts.
  • B Cells: Researchers commonly examine how ALCAM (CD166 antigen) relates to this theme using model systems and orthogonal readouts.
  • Immunology: Researchers commonly examine how ALCAM (CD166 antigen) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative ALCAM (CD166 antigen) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of ALCAM (CD166 antigen) across tissue regions and cell types using matched controls.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
