  • Antibody format: Monoclonal; clone AFBD-3; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

CD2 (protein: T-cell surface glycoprotein CD4) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Cleaves the membrane-bound precursor of TNF-alpha at '76-Ala-|-Val-77' to its mature soluble form. Responsible for the proteolytical release of soluble JAM3 from endothelial cells surface. Responsible for the proteolytic release of several other cell-surface proteins, including heparin-binding epidermal growth- like factor, ephrin-A2 and for constitutive and regulated alpha- secretase cleavage of amyloid precursor protein (APP). Contributes to the normal cleavage of the cellular prion protein. Involved in the cleavage of the adhesion molecule L1 at the cell surface and in released membrane vesicles, suggesting a vesicle-based protease activity. Controls also the proteolytic processing of Notch and mediates lateral inhibition during neurogenesis. Responsible for the FasL ectodomain shedding and for the generation of the remnant ADAM10-processed FasL (FasL APL) transmembrane form. Also cleaves the ectodomain of the integral membrane proteins CORIN and ITM2B. May regulate the EFNA5-EPHA3 signaling. . Reported cellular localization context: Cell membrane; Single-pass type I membrane protein. Endomembrane system; Single-pass type I membrane protein. Is localized in the plasma membrane but is predominantly expressed in the Golgi apparatus and in released membrane vesicles derived likely from the Golgi. Tissue expression notes (as provided): Expressed in spleen, lymph node, thymus, peripheral blood leukocyte, bone marrow, cartilage, chondrocytes and fetal liver. .

Research relevance and current trends

  • Research context keywords from the source record include: Adaptive Immunity,Hematopoietic Progenitors,Immunology,Lymphoid,Myeloid,Stem Cells,T Cells,T Lymphocytic Lineage.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.
  • Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.

Workflow ideas (metafield): Validate CD2 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect CD2 expression by Western blot in cell or tissue lysates, Detect CD2 in FFPE tissue sections by immunohistochemistry, Localize CD2 by immunofluorescence/immunocytochemistry in cultured cells, Quantify CD2-positive cells by flow cytometry in single-cell suspensions, Enrich CD2 by immunoprecipitation from lysates for downstream analysis

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 26 kDa; calculated MW: 84142 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 26 kDa
  • Cellular localization (provided): Cell membrane; Single-pass type I membrane protein. Endomembrane system; Single-pass type I membrane protein. Is localized in the plasma membrane but is predominantly expressed in the Golgi apparatus and in released membrane vesicles derived likely from the Golgi.
  • Tissue details (provided): Expressed in spleen, lymph node, thymus, peripheral blood leukocyte, bone marrow, cartilage, chondrocytes and fetal liver. .

Our lab used your anti-CD2 Monoclonal antibody for WB on leukocyte a few years ago. I am using human, and We are going to use the antibody for IF next. My lab would like examining leukocyte as well as pancreas spleen in our next experiment. Could give a recommendation on which antibody would work the best for IF?
I looked at the website and datasheets of our anti-CD2 Monoclonal antibody and it appears that M00570 has been validated on human in both WB and IF. Thus M00570 should work for your application. Our Boster satisfaction guarantee will cover this product for IF in human even if the specific tissue type has not been validated. We do have a comprehensive range of products for IF detection and you can check out our website bosterbio.com to find out more information about them.
I was wanting to use your anti-CD2 Monoclonal antibody for IP for human leukocyte on frozen tissues, but I want to know if it has been validated for this particular application. Has this antibody been validated and is this antibody a good choice for human leukocyte identification?
It shows on the product datasheet, M00570 anti-CD2 Monoclonal antibody has been validated for IP, IF, WB on human tissues. We have an innovator award program that if you test this antibody and show it works in human leukocyte in IHC-frozen, you can get your next antibody for free.
