Product Overview

Clone FHB-3 is a rabbit monoclonal antibody that recognizes complement receptor type 2 (gene CR2) in human and mouse samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 112.9 kDa, and the supplier reports an observed band at about 155 kDa.

Complement receptor 2 (CR2, CD21) is expressed on B cells and follicular dendritic cells and binds C3d on complement-tagged antigens. As part of the CD19–CD21–CD81 co-receptor complex it lowers the threshold for B-cell activation, and it is also the receptor used by Epstein-Barr virus to enter B cells.

Samples with confirmed staining (IHC): human spleen tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human CD21.

Reactivity Notes

The supplier lists reactivity with human and mouse. UniProt places complement receptor type 2 in the cell membrane. Tissue expression noted by UniProt (human): Mature B-lymphocytes, T-lymphocytes, pharyngeal epithelial cells, astrocytes and follicular dendritic cells of the spleen. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:1000–1:5000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 112.9 kDa; observed band at about 155 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human CD21.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
