Vendor description (summary): Boster Bio Anti-CD22 Antibody Picoband® (monoclonal, 5E7) catalog # M01572-2.

Biological background

Biological context: Mediates B-cell B-cell interactions. May be involved in the localization of B-cells in lymphoid tissues. Binds sialylated glycoproteins; one of which is CD45. Preferentially binds to alpha-2,6-linked sialic acid. The sialic acid recognition site can be masked by cis interactions with sialic acids on the same cell surface. Upon ligand induced tyrosine phosphorylation in the immune response seems to be involved in regulation of B-cell antigen receptor signaling. Plays a role in positive regulation through interaction with Src family tyrosine kinases and may also act as an inhibitory receptor by recruiting cytoplasmic phosphatases via their SH2 domains that block signal transduction through dephosphorylation of signaling molecules.

Expression and localization notes: cellular localization: Cell membrane; Single-pass type I membrane protein, tissue context: B-lymphocytes..

Common research applications

  • Western blotting (WB): Compare CD22 levels across samples and conditions using appropriate loading and biological controls.
  • Immunohistochemistry (IHC): Evaluate spatial distribution of CD22 in tissue sections, considering fixation and antigen retrieval effects.
  • Flow cytometry: Quantify CD22-positive populations in single-cell suspensions with appropriate gating and controls.
  • ELISA: Use antibody-based detection formats to assess antigen presence or binding in plate-based assays.

Notes for experimental interpretation

  • Account for isoforms, post-translational modifications, and sample-specific processing that can shift apparent molecular weight or epitope accessibility.
  • Use positive/negative biological controls where possible (e.g., known-expressing cells/tissues, knockdown/knockout models) and include appropriate secondary-only/isotype controls for imaging workflows.

Additional product notes (from provided fields)

  • Specificity: No cross reactivity with other proteins.
  • Background: CD22 is a surface glycoprotein of B lymphocytes that is rapidly phosphorylated on cytoplasmic tyrosines after antigen receptor cross-linking. It is a negative regulator of antigen receptor signaling whose onset of expression at the mature B cell stage may serve to raise the antigen concentration threshold required for B cell triggering. The human CD22 gene is expressed specifically in B lymphocytes and likely has an important function in cell-cell interactions. The B cell coreceptor CD22 plays an important role in regulating signal transduction via the B cell Ag receptor. And CD22 is located within the band region q13.1 of chromosome 19.
  • Cross reactivity: No cross-reactivity with other proteins.
  • Cellular localization: Cell membrane; Single-pass type I membrane protein
  • Tissue details: B-lymphocytes.
  • Research category: Actin Binding Proteins,Actin, etc.,Cytoskeleton,Cytoskeleton/ECM,Microfilaments,Signal Transduction
