Product Overview
This rabbit polyclonal antibody recognizes CD226 antigen in human and mouse samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 38.6 kDa, and the supplier reports an observed band at about 70 kDa.
CD226, also called DNAM-1, is an activating receptor on NK cells, cytotoxic T cells and other lymphocytes. It binds CD155 (PVR) and CD112 (nectin-2), promoting adhesion, cytotoxicity and cytokine secretion, and it is studied alongside TIGIT in tumor immunity and immune checkpoint research.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human Jurkat cells, human HT1080 cells, human A549 cells, human HEL cells, mouse thymus tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): Jurkat cells.
Immunogen
Recombinant human CD226 fragment (Glu19–Ala254), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt places CD226 antigen in the cell membrane. Tissue expression noted by UniProt (human): Expressed by peripheral blood T-lymphocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
