Product Overview
This rabbit polyclonal antibody detects tumor necrosis factor ligand superfamily member 4 (gene Tnfsf4) in mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 22.3 kDa, and the supplier reports an observed band at about 21–25 kDa.
OX40 ligand (OX40L, TNFSF4, CD252) is a TNF family cytokine expressed mainly on activated antigen-presenting cells. By binding OX40 (TNFRSF4) on T cells, it costimulates their proliferation, survival and cytokine output, so the OX40L–OX40 pair is studied in allergy, autoimmunity and immunotherapy research.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
Conditions in the example images (WB): 5–20% gradient SDS-PAGE, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): rat small intestine tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Immunogen
Recombinant mouse CD252 fragment (Gln49–Leu198), expressed in E. coli. Sequence identity with the human and rat orthologs: 42.7% and 78.8%, respectively.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places tumor necrosis factor ligand superfamily member 4 in the membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
