Product Overview
This rabbit polyclonal antibody recognizes T-cell-specific surface glycoprotein CD28 in human samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 25.1 kDa, and the supplier reports an observed band at about 60 kDa.
CD28 is the main co-stimulatory receptor on T cells. When it binds CD80 or CD86 on antigen-presenting cells, it amplifies T-cell receptor signals to promote IL-2 production, proliferation and survival, and it is a core molecule in T-cell activation studies and CAR design.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human Jurkat cells, human HUT-78 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): HEL cells.
Immunogen
Recombinant human CD28 fragment (Gln15–Asp214), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places T-cell-specific surface glycoprotein CD28 in the cell membrane and SUBCELLULAR LOCATION:. Tissue expression noted by UniProt (human): Expressed in T-cells and plasma cells, but not in less mature B-cells. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
