Key elements and design rationale

  • Target: CD3E (T-cell surface glycoprotein CD3 epsilon chain).
  • Antibody format: Monoclonal; clone COB-3; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

CD3E (protein: T-cell surface glycoprotein CD3 zeta chain) is a commonly studied target in molecular and cellular biology. Functional context (as provided): The CD3 complex mediates signal transduction. Reported cellular localization context: Membrane; Single-pass type I membrane protein. Tissue expression notes (as provided): Expressed in monocytes, basophils, B-cells, umbilical vein endothelial cells (HUVEC) and B-lymphoblastoid cells. Lower expression detected in CD4+ T-lymphocytes and natural killer cells. In the brain, detected in endothelial cells and capillaries, and in mature neurons of the frontal cortex and hippocampus. Expressed in tubular formation in the kidney. Highly expressed in astroglial tumor endothelial, microglial and glioma cells. Expressed at low levels in normal CD34+ progenitor cells, but at very high levels in several myeloid malignant cell lines. Expressed in breast carcinomas but not in normal breast tissue (at protein level). .

Research relevance and current trends

  • Research context keywords from the source record include: Adaptive Immunity,Hematopoietic Progenitors,Immunology,Stem Cells,T Cells.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Immunohistochemistry (IHC): evaluate spatial distribution of target-positive staining in tissue architecture.
  • Immunofluorescence/ICC (IF/ICC): visualize subcellular localization patterns and cell-to-cell heterogeneity.
  • Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.

Workflow ideas (metafield): Validate CD3E antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect CD3E in FFPE tissue sections by immunohistochemistry, Localize CD3E by immunofluorescence/immunocytochemistry in cultured cells, Enrich CD3E by immunoprecipitation from lysates for downstream analysis

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 250 kDa).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 250 kDa
  • Cellular localization (provided): Membrane; Single-pass type I membrane protein.

Would anti-CD3 epsilon Rabbit Monoclonal antibody M02675-1 work for IHC with blood?
According to the expression profile of blood, CD3E is highly expressed in blood. So, it is likely that anti-CD3 epsilon Rabbit Monoclonal antibody M02675-1 will work for IHC with blood.
Can you help my question with product M02675-1, anti-CD3 epsilon Rabbit Monoclonal antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?
It is not recommended storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free M02675-1 anti-CD3 epsilon Rabbit Monoclonal antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
