Product Overview

This rabbit polyclonal antibody recognizes CMRF35-like molecule 8 (gene CD300A) in human, mouse and rat samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 33.2 kDa, and the supplier reports an observed band at about 60 kDa.

CD300a is an inhibitory immunoglobulin-like receptor expressed on NK cells, mast cells, granulocytes and other leukocytes. It recognizes phosphatidylserine and phosphatidylethanolamine exposed on dying cells and uses ITIM motifs to dampen cell activation, so it is studied in the regulation of inflammation and cytotoxicity.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human, Mouse, Rat)
Flow cytometry (fixed cells) 1–3 µg/1×106 cells (Human)
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human HL-60 cells, human Raji cells, rat PC-12 cells, rat C6 cells, mouse RAW264.7 cells, mouse J774A1 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with a confirmed signal (flow cytometry): U2OS cells.

Immunogen

Recombinant human CD300A fragment (Leu156–Thr299), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 54.7% and 55.7%, respectively.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places CMRF35-like molecule 8 in the cell membrane. Tissue expression noted by UniProt (human): Expressed not only by natural killer (NK) cells but also by T-cell subsets, B-cells, dendritic cells, mast cells, granulocytes and monocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human, Mouse, Rat); Flow cytometry (fixed cells) 1–3 µg/1×10⁶ cells (Human); ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 33.2 kDa; observed band at about 60 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human CD300A fragment (Leu156–Thr299), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 54.7% and 55.7%, respectively.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
