Product Overview
This rabbit polyclonal antibody detects epithelial cell adhesion molecule (gene Epcam) in mouse and rat samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 35.0 kDa, and the supplier reports an observed band at about 38 kDa.
EpCAM (CD326) is a transmembrane glycoprotein on most epithelial cells that mediates homophilic cell-cell adhesion. It also influences proliferation and stem cell maintenance, and it is highly expressed in many carcinomas. EpCAM is widely used to identify and isolate epithelial and circulating tumor cells, and EPCAM mutations cause congenital tufting enteropathy.

|---|---|
| IHC (paraffin sections) | 1–2 µg/mL (Mouse, Rat) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Mouse, Rat) |

Samples with a confirmed band (WB): mouse small intestine tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.25 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): rat colon tissue, mouse colon tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): mouse colon cancer tissue, rat colon cancer tissue.
Samples with a confirmed signal (flow cytometry): RAW264.7 cells.
Immunogen
Recombinant mouse CD326/Epcam fragment (Gln24–Ala267), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt places epithelial cell adhesion molecule in the lateral cell membrane and cell junction. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
