Myeloid cell surface antigen CD33; Myeloid cell surface antigen CD33. CD33, also known as Siglec-3 (sialic acid binding Ig-like lectin 3, SIGLEC3, SIGLEC-3, gp67, p67), is a transmembrane receptor expressed on cells of myeloid lineage. It is usually considered myeloid-specific, but it can also be found on some lymphoid cells. CD33 binds sialic acids, therefore is a member of the SIGLEC family of lectins. By fluorescence in situ hybridization, CD33 is mapped to 19q13.3-q13.4. Functional note: Putative adhesion molecule of myelomonocytic-derived cells that mediates sialic-acid dependent binding to cells. Preferentially binds to alpha-2,6-linked sialic acid (By similarity). The sialic acid recognition site may be masked by cis interactions with sialic acids on the same cell surface. . Reported localization: Cell membrane; Single-pass type I membrane protein. Will anti-CD33 antibody RP1105 work for IHC with premonocytic lymphoma?
According to the expression profile of premonocytic lymphoma, CD33 is highly expressed in premonocytic lymphoma. So, it is likely that anti-CD33 antibody RP1105 will work for IHC with premonocytic lymphoma.
My question regarding product RP1105, anti-CD33 antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?
We suggest not storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free RP1105 anti-CD33 antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
