Product Overview

Clone IDE-3 is a rabbit monoclonal antibody that recognizes T-cell surface glycoprotein CD3 delta chain (gene CD3D) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 18.9 kDa, and the supplier reports an observed band at about 21 kDa.

CD3δ (CD3D) is one of the invariant chains of the T-cell receptor–CD3 complex, pairing with CD3ε. Its ITAM motif transmits signals after antigen recognition, and it is needed for normal T-cell development in the thymus.

Samples with a confirmed band (WB): human Jurkat cells, human MOLT-4 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.

Samples with confirmed staining (IHC): human spleen tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Samples with confirmed staining (ICC/IF): Jurkat cells.

Immunogen

Synthetic peptide based on the sequence of human CD3D.

Reactivity Notes

The supplier lists reactivity with human. UniProt places T-cell surface glycoprotein CD3 delta chain in the cell membrane. Tissue expression noted by UniProt (human): CD3D is mostly present on T-lymphocytes with its TCR-CD3 partners. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:20; Immunoprecipitation 1:20. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 18.9 kDa; observed band at about 21 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human CD3D.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
