Product Overview
This rabbit polyclonal antibody detects receptor-type tyrosine-protein phosphatase C (gene PTPRC) in human samples and is validated for flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 147.5 kDa, and the supplier reports an observed band at about 180–250 kDa.
CD45 (PTPRC) is a transmembrane tyrosine phosphatase present on all nucleated blood cells. By removing inhibitory phosphates from Src family kinases such as LCK and FYN it enables T- and B-cell antigen receptor signaling, and its isoforms (for example CD45RA and CD45RO) are widely used to mark leukocyte subsets.

Samples with a confirmed band (WB): human Jurkat cells, human Raji cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): human tonsil tissue.
Samples with a confirmed signal (flow cytometry): Jurkat cells.
Immunogen
Synthetic peptide from the C-terminal region of human CD45. It differs from the mouse sequence at eight residues and from the rat sequence at ten residues.
Reactivity Notes
The supplier lists reactivity with human. UniProt places receptor-type tyrosine-protein phosphatase C in the cell membrane, membrane raft and synapse. Tissue expression noted by UniProt (human): Isoform 1: Detected in thymocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
