Product Overview
Clone AEI-3 is a rabbit monoclonal antibody that recognizes membrane cofactor protein (gene CD46) in human samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 43.7 kDa, and the supplier reports an observed band at about 70 kDa.
CD46, or membrane cofactor protein, is a widely expressed complement regulator that serves as a cofactor for factor I in cleaving C3b and C4b on host cells. It protects self tissue from complement attack, also regulates T-cell responses, and is used as a receptor by measles virus and several other pathogens.

Samples with a confirmed band (WB): human HeLa cells, human HepG2 cells, human RT4 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon tissue, human colon cancer tissue, human prostate cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

The supplier lists reactivity with human. Tissue expression noted by UniProt (human): Expressed by all cells except erythrocytes. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
