Anti-CD63 (extracellular) Antibody (#ANR-210) is a highly specific antibody directed against an extracellular epitope of the mouse protein. The antibody can be used in western blot, immunohistochemistry and flow cytometry applications. It has been designed to recognize CD63 from mouse and rat samples. Anti-CD63 (extracellular)-PE Antibody (#ANR-210-PE) is directly conjugated to R-Phycoerythrin (R-PE) fluorophore. This conjugated antibody has been developed to be used in immunofluorescent applications such as direct flow cytometry and live cell imaging. The antibody will not work with human samples.

Target CD63

Overview

Anti-CD63 (extracellular)-PE Antibody is an antibody targeting Tetraspanin-30, TSPAN30, Lysosomal-Associated Membrane Protein 3, LAMP-3, Granulophysin, Melanoma-Associated Antigen ME491 Polyclonal raised in Rabbit (R-Phycoerythrin (R-PE)). This antibody is commonly used in FC, LCI to detect, localize, or compare expression of the target across samples.

Key elements and design rationale

  • Target: Tetraspanin-30, TSPAN30, Lysosomal-Associated Membrane Protein 3, LAMP-3, Granulophysin, Melanoma-Associated Antigen ME491 (also reported as Tetraspanin-30, TSPAN30, Lysosomal-Associated Membrane Protein 3, LAMP-3, Granulophysin, Melanoma-Associated Antigen ME491).
  • Immunogen/epitope region: Extracellular, 2nd loop..
  • Homology note: Rat - identical Won't recognize CD63 from human samples (informative for cross-species interpretation).
  • Species reactivity (as provided): Rat, Mouse.
  • Lot quality control (as provided): Western blot analysis (unlabeled antibody, #ANR-210), and direct flow cytometry (labeled antibody)..
  • Peptide confirmation: Confirmed by amino acid analysis and mass spectrometry.
  • Blocking peptide: Available for antigen preadsorption control where appropriate.
  • Conjugate/format: R-Phycoerythrin (R-PE) (may affect detection channel and background).

These attributes help researchers interpret whether signal reflects the intended target in a given assay and sample context.

Biological background

CD63, also known as Tetraspanin-30 (Tspan-30), lysosomal-associated membrane protein 3 (LAMP-3), lysosome integral membrane protein 1 (Limp1), melanoma-associated antigen (ME491), and ocular melanoma-associated antigen (OMA81H), is a multi-transmembrane protein that plays a role in cancer malignancy.1CD63 is a highly N-glycosylated member of the transmembrane-4 superfamily of tetraspanin proteins. CD63 is composed of 238 amino acids comprising four transmembrane α-helices, two extracellular loops, and a short cytoplasmic tail. CD63 is located in late endosomes, lysosomes, secretory vesicles, and the plasma membrane.

Research relevance and current trends

  • Profiling immune-marker expression across cell subsets with single-cell or flow-based readouts.
  • Connecting receptor/ligand levels to activation state and cytokine programs.
  • Applying genetic perturbation or orthogonal assays to support specificity and interpretation.

Common research applications

  • Flow cytometry (direct/indirect): quantify target-positive populations and shifts in expression across subsets.
  • Live cell imaging (LCI): support extracellular-epitope detection on non-permeabilized cells when appropriate.

Interpretation typically benefits from comparing matched sample sets (e.g., treated vs control, WT vs KO/KD) and using orthogonal readouts where feasible.

Notes for experimental interpretation

  • Isoforms and post-translational modifications can shift apparent molecular weight or epitope accessibility across samples.
  • Cross-species signal may depend on epitope conservation; consult the provided homology note when selecting models.
  • Permeabilization, fixation, and antigen retrieval can change accessibility of intracellular vs extracellular epitopes.
  • Conceptual control: antigen preadsorption (blocking peptide) can help assess signal dependence on the immunogen region.
  • Provided control suggestions: Negative control: RIC-001-PE.
  • Application notes: see product-specific dilution/usage notes and control concepts provided in the dataset.

Application abbreviations: CBE- Cell-based ELISA, FC- Flow cytometry, ICC- Immunocytochemistry, IE- Indirect ELISA, IF- Immunofluorescence, IFC- Indirect flow cytometry, IHC- Immunohistochemistry, IP- Immunoprecipitation, LCI- Live cell imaging, N- Neutralization, WB- Western blot. Species abbreviations: H- Human, M- Mouse, R- Rat.
