Product Overview
This rabbit polyclonal antibody detects HLA class II histocompatibility antigen gamma chain (gene CD74) in human samples and is validated for Western blot, IHC, ICC/IF, flow cytometry and ELISA. The predicted molecular weight is 33.5 kDa, and the supplier reports an observed band at about 31–34 kDa.
CD74, the MHC class II invariant chain, binds newly made class II α/β heterodimers in the endoplasmic reticulum and guides them to endosomal compartments for peptide loading. It is also a cell surface receptor for macrophage migration inhibitory factor (MIF), linking it to cell survival and inflammatory signaling.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human Daudi cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human spleen tissue, human tonsil tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): human spleen tissue.
Samples with a confirmed signal (flow cytometry): Daudi cells.
Immunogen
Recombinant human CD74 fragment (Gln73–Met296), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human. UniProt places HLA class II histocompatibility antigen gamma chain in the cell membrane, endoplasmic reticulum membrane, Golgi apparatus and endosome. Tissue expression noted by UniProt (human): Detected in urine (at protein level)... The samples tested by the supplier (listed above) are a practical starting point for positive controls.
