Overview

This antibody is intended for detection of CD8A (T-cell surface glycoprotein CD8 alpha chain) in biological samples using common immunoassay formats. It is typically selected based on target identity, species reactivity, clonality/clone information, and detection modality.

Vendor notes: Boster Bio Anti-CD8 alpha/CD8A Antibody Picoband® catalog # PB9249. Tested in IHC, WB applications. This antibody reacts with Human. The brand Picoband indicates this is a premium antibody that guarantees superior quality, high affinity, and strong signals with minimal background in Western blot applications. Only our best-performing antibodies are designated as Picoband, ensuring unmatched performance.

Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E.coli-derived human CD8 alpha recombinant protein (Position: S22-A187). Human CD8 alpha shares 49% and 47% amino acid (aa) sequence identity with mouse and rat CD8 alpha, respectively. (reported region: S22-A187).
  • Molecular weight context: reported MW: 55 kDa; calculated MW: 25729 MW
  • Reactivity: Human
  • Applications: IHC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

T-cell surface glycoprotein CD8 alpha chain; T-cell surface glycoprotein CD8 alpha chain. The CD8 antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes that mediates efficient cell-cell interactions within the immune system. It is mapped to 2p11.2. The CD8 antigen, acting as a coreceptor, and the T-cell receptor on the T lymphocyte recognize antigen displayed by an antigen presenting cell (APC) in the context of class I MHC molecules. The functional coreceptor is either a homodimer composed of two alpha chains, or a heterodimer composed of one alpha and one beta chain. Both alpha and beta chains share significant homology to immunoglobulin variable light chains. This gene also encodes the CD8 alpha chain isoforms. Functional note: Identifies cytotoxic/suppressor T-cells that interact with MHC class I bearing targets. CD8 is thought to play a role in the process of T-cell mediated killing. CD8 alpha chains binds to class I MHC molecules alpha-3 domains. Reported localization: Isoform 1: Cell membrane; Single-pass type I membrane protein. Expression/tissue context: Brain.

Research relevance and current trends

  • Adaptive Immunity: Researchers commonly examine how CD8A (T-cell surface glycoprotein CD8 alpha chain) relates to this theme using model systems and orthogonal readouts.
  • Immunology: Researchers commonly examine how CD8A (T-cell surface glycoprotein CD8 alpha chain) relates to this theme using model systems and orthogonal readouts.
  • T Cells: Researchers commonly examine how CD8A (T-cell surface glycoprotein CD8 alpha chain) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative CD8A (T-cell surface glycoprotein CD8 alpha chain) levels across conditions; band patterns may reflect isoforms and processing.
  • IHC/IHC-F: assess spatial distribution of CD8A (T-cell surface glycoprotein CD8 alpha chain) across tissue regions and cell types using matched controls.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Family / similarity context: Contains 1 Ig-like V-type (immunoglobulin-like) domain.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
