Biological background

Protein function (datasheet): Plasma membrane-associated small GTPase which cycles between an active GTP-bound and an inactive GDP-bound state. In active state binds to a variety of effector proteins to regulate cellular responses. Involved in epithelial cell polarization processes. Regulates the bipolar attachment of spindle microtubules to kinetochores before chromosome congression in metaphase. Plays a role in the extension and maintenance of the formation of thin, actin-rich surface projections called filopodia. Mediates CDC42-dependent cell migration. .

Cellular localization (datasheet): Cell membrane ; Lipid-anchor ; Cytoplasmic side . Cytoplasm, cytoskeleton, microtubule organizing center, centrosome. Cytoplasm, cytoskeleton, spindle. Midbody. Localizes to spindle during prometaphase cells. Moves to the central spindle as cells progressed through anaphase to telophase. Localizes at the end of cytokinesis in the intercellular bridge formed between two daughter cells. Its localization is regulated by the activities of guanine nucleotide exchange factor ECT2 and GTPase activating protein RACGAP1. Colocalizes with NEK6 in the centrosome.

Tissue details (datasheet): Highly expressed in heart, placenta, skeletal muscle, kidney, pancreas, spleen, thymus, prostate, testis and peripheral blood.

Research relevance and current trends

  • Commonly studied in contexts related to Cancer,Cell Biology,Cell Cycle,Cell Division,Cytokinesis,Epigenetics and Nuclear Signaling,G Protein Signaling,Kinases/Phosphatases,Protein Trafficking,Ras Family,Signal Transduction,Signaling Pathway,Small G Proteins,Vesicle Transport.
  • Supports comparative expression analysis across conditions, genotypes, or treatments when paired with appropriate controls.
  • Useful for confirming target presence and subcellular distribution using orthogonal readouts (e.g., microscopy vs. immunoblotting).

Common research applications

  • Western blot (WB): Compare relative target abundance and apparent size/isoforms across samples; interpret bands in light of expected MW and potential PTMs.
  • Immunohistochemistry (IHC): Assess tissue distribution and cell-type patterns; interpret staining with appropriate negative controls and antigen context.
  • Flow cytometry: Quantify target-positive populations in single-cell suspensions; pair with viability and isotype/FMO controls conceptually.

Notes for experimental interpretation

  • Consider isoforms, post-translational modifications, and processing that can shift apparent molecular weight or localization.
  • Use appropriate positive and negative controls (e.g., KO/KD, blocking peptide, or isotype controls) to support specificity interpretation.

As a monoclonal antibody, this reagent is expected to recognize a defined epitope, which can support consistency across lots when epitope accessibility is preserved.

We are currently using anti-CDC42 Rabbit Monoclonal antibody M00119 for rat tissue, and we are content with the IP results. The species of reactivity given in the datasheet says human, mouse, rat. Is it possible that the antibody can work on primate tissues as well?
The anti-CDC42 Rabbit Monoclonal antibody (M00119) has not been validated for cross reactivity specifically with primate tissues, but there is a good chance of cross reactivity. We have an innovator award program that if you test this antibody and show it works in primate you can get your next antibody for free. Please contact me if I can help you with anything.
We have tried in the past anti-CDC42 Rabbit Monoclonal antibody for IP on fetal brain a few months ago. I am using human, and I plan to use the antibody for WB next. I am looking for examining fetal brain as well as cervix in our next experiment. Could you please give me some suggestion on which antibody would work the best for WB?
I looked at the website and datasheets of our anti-CDC42 Rabbit Monoclonal antibody and I see that M00119 has been tested on human in both IP and WB. Thus M00119 should work for your application. Our Boster satisfaction guarantee will cover this product for WB in human even if the specific tissue type has not been validated. We do have a comprehensive range of products for WB detection and you can check out our website bosterbio.com to find out more information about them.
