Product Overview

Clone 6D5B5 is a mouse monoclonal antibody that recognizes cyclin-dependent kinase 2 (gene CDK2) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 33.9 kDa, and the supplier reports an observed band at about 30 kDa.

CDK2 is a cyclin-dependent kinase that pairs with cyclin E and cyclin A to drive the G1-to-S transition and DNA replication. It phosphorylates targets such as RB1 and helps trigger centrosome duplication; although somatic cells can often compensate for its loss, it is essential for meiosis and is widely studied in cell cycle research.

Validated Applications

Western blot 0.25–0.5 µg/mL (Human, Mouse, Rat)
IHC (paraffin sections) 2–5 µg/mL (Human, Mouse, Rat)

Samples with a confirmed band (WB): human Jurkat cells, human HepG2 cells, human U2OS cells, rat heart tissue, rat L6 cells, mouse heart tissue, mouse C2C12 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Samples with confirmed staining (IHC): human laryngeal squamous cells carcinomas tissue, human liver cancer tissue, human renal clear cells carcinoma tissue, human serous adenocarcinoma of ovary tissue, mouse colon tissue, rat colon tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Recombinant human Cdk2 fragment (Glu81–Leu298), expressed in E. coli. Sequence identity with the rat ortholog: 98.6%.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places cyclin-dependent kinase 2 in the nucleus, cytoplasm and endosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

For research use only; not for diagnostic or therapeutic procedures. Handle with standard laboratory precautions.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.25–0.5 µg/mL (Human, Mouse, Rat); IHC (paraffin sections) 2–5 µg/mL (Human, Mouse, Rat). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 33.9 kDa; observed band at about 30 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Recombinant human Cdk2 fragment (Glu81–Leu298), expressed in E. coli. Sequence identity with the rat ortholog: 98.6%.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
