Product Overview
Clone BO-3 is a rabbit monoclonal antibody that recognizes DNA replication factor Cdt1 in human samples and is validated for Western blot, IHC, ICC/IF and immunoprecipitation. The predicted molecular weight is 60.4 kDa, and the supplier reports an observed band at about 70 kDa.
Cdt1 is a DNA replication licensing factor that works with ORC and CDC6 during G1 to load the MCM helicase onto replication origins. Its activity is restricted by geminin and by degradation after S phase begins, which prevents re-replication; it also contributes to kinetochore-microtubule attachment during mitosis.

Samples with a confirmed band (WB): human 293T cells, human K562 cells, human Jurkat cells, human PC-3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
