Product Overview
Clone AAF-3 is a rabbit monoclonal antibody that recognizes serine/threonine-protein kinase Chk1 (gene CHEK1) in human, mouse and rat samples and is validated for Western blot, ICC/IF and immunoprecipitation. The predicted molecular weight is 54.4 kDa, and the supplier reports an observed band at about 54 kDa.
Chk1 (CHEK1) is a serine/threonine kinase that enforces DNA damage and replication checkpoints. Activated by ATR, it phosphorylates CDC25 phosphatases to block CDK activation and pause the cell cycle while DNA is repaired, making it central to genome maintenance and a target in cancer research.

Samples with a confirmed band (WB): human K562 cells, human Caco-2 cells, human Jurkat cells, human 293T cells, rat PC-12 cells, mouse thymus tissue, mouse SP2/0 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Immunogen
Synthetic peptide based on the sequence of human chk1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places serine/threonine-protein kinase Chk1 in the nucleus, chromosome and cytoplasm. Tissue expression noted by UniProt (human): Expressed ubiquitously with the most abundant expression in thymus, testis, small intestine and colon. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
