Product Overview
Clone BBC-3 is a rabbit monoclonal antibody that recognizes casein kinase II subunit alpha (gene CSNK2A1) in human, mouse and rat samples and is validated for Western blot, IHC and flow cytometry. The predicted molecular weight is 45.1 kDa, and the supplier reports an observed band at about 40 kDa.
CK2 alpha (CSNK2A1) is a catalytic subunit of casein kinase 2, a constitutively active serine/threonine kinase that phosphorylates hundreds of substrates near acidic residues. CK2 influences cell cycle progression, transcription, DNA repair and apoptosis, including phosphorylation of p53 and caspases, and it is widely studied in cell survival signaling and cancer.

Samples with a confirmed band (WB): human HepG2 cells, human A431 cells, human A549 cells, human AGS cells, rat heart tissue, rat C6 cells, mouse heart tissue, mouse Neuro-2a cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human breast cancer tissue, rat heart tissue, mouse heart tissue, mouse brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places casein kinase II subunit alpha in the nucleus. Tissue expression noted by UniProt (human): Expressed in gastric carcinoma tissue and the expression gradually increases with the progression of the carcinoma (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
