Product Overview
This rabbit polyclonal antibody recognizes claudin-3 (gene CLDN3) in human samples and is validated for Western blot, IHC, ICC/IF and flow cytometry. The predicted molecular weight is 23.3 kDa, and the supplier reports an observed band at about 23 kDa.
Claudin-3 (CLDN3) is a barrier-forming member of the claudin family that assembles into tight junction strands and seals the space between neighboring epithelial cells. It is expressed in many epithelia, and researchers use it as a marker of tight junction organization in studies of epithelial barrier function and cancer biology.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
| Immunocytochemistry / Immunofluorescence | 5 µg/mL (Human) |
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
Samples with a confirmed band (WB): human Caco-2 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): human pancreatic cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Samples with confirmed staining (ICC/IF): MCF-7 cells.
Samples with a confirmed signal (flow cytometry): PC-3 cells.
Immunogen
Synthetic peptide from the C-terminal region of human claudin 3/CLDN3. Sequence identity with the mouse and rat orthologs: 61.1% and 72.2%, respectively.
Reactivity Notes
The supplier lists reactivity with human. UniProt places claudin-3 in the cell junction and cell membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
