Key elements and design rationale

  • Target: CASP9 (Caspase-9).
  • Antibody format: Monoclonal; clone FEE-3; isotype Rabbit IgG.
  • Host: Rabbit.
  • Species reactivity: Human,Mouse (confirm in your model system with appropriate controls).

This description is intended to help interpret the antibody design and the biological context of the target using the fields provided in the catalog record, alongside general experimental considerations.

Biological background

CASP9 (protein: P2X purinoceptor 1) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Involved in the activation cascade of caspases responsible for apoptosis execution. Binding of caspase-9 to Apaf- 1 leads to activation of the protease which then cleaves and activates caspase-3. Promotes DNA damage-induced apoptosis in a ABL1/c-Abl-dependent manner. Proteolytically cleaves poly (ADP- ribose) polymerase (PARP). Reported cellular localization context: Cytoplasm, cytosol. Nucleus. Cytosolic under unstressed conditions, translocates into the nucleus upon induction by electrophilic agents. Tissue expression notes (as provided): Ubiquitous, with highest expression in the heart, moderate expression in liver, skeletal muscle, and pancreas. Low levels in all other tissues. Within the heart, specifically expressed in myocytes. .

Research relevance and current trends

  • Research context keywords from the source record include: Apoptosis,Apoptotic Markers,Cancer,Caspases,Cell Biology,Cell Death,Intracellular,Invasion/Microenvironment,Metabolism,Metabolism Processes,Pathways and Processes.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.

Workflow ideas (metafield): Validate CASP9 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect CASP9 expression by Western blot in cell or tissue lysates, Enrich CASP9 by immunoprecipitation from lysates for downstream analysis

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 46 kDa; calculated MW: 46281 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 46 kDa
  • Cellular localization (provided): Cytoplasm, cytosol. Nucleus. Cytosolic under unstressed conditions, translocates into the nucleus upon induction by electrophilic agents.
  • Tissue details (provided): Ubiquitous, with highest expression in the heart, moderate expression in liver, skeletal muscle, and pancreas. Low levels in all other tissues. Within the heart, specifically expressed in myocytes. .

Would M00080-2 anti-cleaved Caspase-9 Rabbit Monoclonal antibody work on parafin embedded sections? If so, which fixation method do you recommend we use (PFA, paraformaldehyde, other)?
It shows on the product datasheet, M00080-2 anti-cleaved Caspase-9 Rabbit Monoclonal antibody as been validated on IP. It is best to use PFA for fixation because it has better tissue penetration ability. PFA needs to be prepared fresh before use. Long term stored PFA turns into formalin, as the PFA molecules congregate and become formalin.
We bought anti-cleaved Caspase-9 Rabbit Monoclonal antibody for WB on stomach cancer in a previous project. I am using mouse, and We intend to use the antibody for IP next. I am interested in examining stomach cancer as well as adrenal tissue in our next experiment. Could you please give me some suggestion on which antibody would work the best for IP?
I have checked the website and datasheets of our anti-cleaved Caspase-9 Rabbit Monoclonal antibody and it appears that M00080-2 has been validated on mouse in both WB and IP. Thus M00080-2 should work for your application. Our Boster satisfaction guarantee will cover this product for IP in mouse even if the specific tissue type has not been validated. We do have a comprehensive range of products for IP detection and you can check out our website bosterbio.com to find out more information about them.
