Product Overview

Clone 21C89 is a rabbit monoclonal antibody that recognizes calsyntenin-1 (gene CLSTN1) in human and mouse samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 109.8 kDa, and the supplier reports an observed band at about 150 kDa.

Calsyntenin-1 (CLSTN1) is a transmembrane cadherin-superfamily protein enriched in neurons. It acts as a synaptic adhesion molecule that binds presynaptic neurexins to support synapse formation, and it also serves as a kinesin-1 cargo adaptor in axonal transport, so it is studied in synapse development and neuronal trafficking.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200

Samples with a confirmed band (WB): 293T cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): mouse brain tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide based on the sequence of human CLSTN1.

Reactivity Notes

The supplier lists reactivity with human and mouse. UniProt places calsyntenin-1 in the postsynaptic cell membrane, endoplasmic reticulum membrane, Golgi apparatus membrane and cell projection. Tissue expression noted by UniProt (human): Expressed in the brain and, a lower level, in the heart, skeletal muscle, kidney and placenta. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 109.8 kDa; observed band at about 150 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human CLSTN1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
