Product Overview

Clone 17C92 is a rabbit monoclonal antibody that recognizes clusterin in human and mouse samples and is validated for Western blot and IHC. The predicted molecular weight is 52.5 kDa, and the supplier reports an observed band at about 65 kDa.

Clusterin (CLU, also called apolipoprotein J) is a secreted glycoprotein that works as an extracellular chaperone, binding misfolded proteins and preventing their aggregation, including amyloid-forming proteins. It is abundant in plasma and many body fluids, and it is widely studied in protein homeostasis, lipid transport, neurodegeneration and cell survival.

Validated Applications

Western blot 1:500–1:2000
Immunohistochemistry 1:50–1:200

Samples with a confirmed band (WB): human HeLa cells, human U2OS cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human clusterin.

Reactivity Notes

The supplier lists reactivity with human and mouse. Tissue expression noted by UniProt (human): Detected in blood plasma, cerebrospinal fluid, milk, seminal plasma and colon mucosa. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 52.5 kDa; observed band at about 65 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human clusterin.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
