Product Overview

This rabbit polyclonal antibody recognizes ciliary neurotrophic factor (gene Cntf) in mouse and rat samples and is validated for Western blot, IHC and ELISA. The predicted molecular weight is 22.9 kDa, and the supplier reports an observed band at about 23 kDa.

Ciliary neurotrophic factor (CNTF) is a cytokine of the IL-6 family that supports the survival of neurons and glia. It signals through a receptor complex of CNTFRα, gp130 and LIFR, activating JAK–STAT pathways, and it is studied in neuroprotection, retinal degeneration and nerve injury.

Samples with a confirmed band (WB): rat PC-12 cells, rat NRK cells, rat RH-35 cells, rat brain tissue, rat liver tissue, rat spleen tissue, rat testis tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Samples with confirmed staining (IHC): mouse brain tissue, rat brain tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Recombinant rat CNTF fragment (Ala2–Met200), expressed in E. coli. Sequence identity with the human and mouse orthologs: 84% and 95%, respectively.

Reactivity Notes

The supplier lists reactivity with mouse and rat. UniProt describes ciliary neurotrophic factor as secreted and also located in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
