Product Overview
This rabbit polyclonal antibody targets collagen alpha-1(I) chain (gene COL1A1) in human and mouse samples and is validated for Western blot and IHC. The predicted molecular weight is 138.9 kDa, and the supplier reports an observed band at about 180–200 kDa.
Collagen α1(I) chain (COL1A1) is the major chain of type I collagen, the most abundant fibrillar collagen in bone, skin, tendon and other connective tissues. It is a standard marker of fibroblasts, osteoblasts and fibrosis, and COL1A1 mutations cause osteogenesis imperfecta.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse) |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL (Human) |
Samples with a confirmed band (WB): human HK2 cells, human HCC tissue, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human colon adenocarcinoma tissue, human gastric carcinomaa tissue, human spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the C-terminal region of human collagen I. It differs from the rat and mouse sequences at two residues.
Reactivity Notes
The supplier lists reactivity with human and mouse. Tissue expression noted by UniProt (human): Forms the fibrils of tendon, ligaments and bones. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
