Product Overview
This rabbit polyclonal antibody detects collagen alpha-1(II) chain (gene COL2A1) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 141.8 kDa, and the supplier reports an observed band at about 180–190 kDa.
Collagen α1(II) chain (COL2A1) forms type II collagen, the main collagen of hyaline cartilage and the vitreous of the eye. It is essential for skeletal development and for cartilage resistance to compression, and it is a standard chondrocyte marker in cartilage and osteoarthritis research.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|
Samples with a confirmed band (WB): rat cartilage tissue, mouse cartilage tissue, mouse auricular cartilage.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Immunogen
Synthetic peptide from the C-terminal region of human collagen II. Sequence identity with the mouse and rat orthologs: 91.7% and 95.8%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Isoform 2 is highly expressed in juvenile chondrocyte and low in fetal chondrocyte. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
