Product Overview

This rabbit polyclonal antibody recognizes collagen alpha-1(II) chain (gene Col2a1) in mouse and rat samples and is validated for Western blot, IHC and ICC/IF. The predicted molecular weight is 142.0 kDa, and the supplier reports an observed band at about 170 kDa.

Collagen α1(II) chain (COL2A1) forms type II collagen, the main collagen of hyaline cartilage and the vitreous of the eye. It is essential for skeletal development and for cartilage resistance to compression, and it is a standard chondrocyte marker in cartilage and osteoarthritis research.

Validated Applications

Western blot 0.1–0.5 µg/mL (Rat, Mouse)
IHC (frozen sections) 0.5–1 µg/mL (Rat, Mouse)
Immunocytochemistry / Immunofluorescence Not specified by the supplier

Samples with a confirmed band (WB): rat trachea tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (ICC/IF): mouse cartilage tissue.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Synthetic peptide from the C-terminal region of mouse collagen II1222–1241aa AFAGLGQREKGPDPMQYMRA). It differs from the rat sequence at one residue.

Reactivity Notes

The supplier lists reactivity with mouse and rat. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Rat, Mouse); IHC (frozen sections) 0.5–1 µg/mL (Rat, Mouse); Immunocytochemistry / Immunofluorescence (dilution not specified). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on zebrafish samples?
A.Reactivity with zebrafish samples has not been tested by the supplier. Aligning the immunogen sequence with the zebrafish ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 142.0 kDa; observed band at about 170 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of mouse collagen II1222–1241aa AFAGLGQREKGPDPMQYMRA). It differs from the rat sequence at one residue.
