Product Overview
This rabbit polyclonal antibody recognizes cartilage oligomeric matrix protein (gene COMP) in human, mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 82.9 kDa, and the supplier reports an observed band at about 110 kDa.
Cartilage oligomeric matrix protein (COMP, thrombospondin-5) is a pentameric extracellular matrix glycoprotein abundant in cartilage, tendon and ligament. It binds collagens and fibronectin to stabilize the matrix, mutations cause pseudoachondroplasia, and COMP is often measured as a marker of cartilage turnover in arthritis research.

Samples with a confirmed band (WB): human HepG2 cells, human Hacat cells, human Caco-2 cells, human 293T cells, rat cartilage tissue, mouse cartilage tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse knee-joint tissue, rat knee-joint tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Recombinant human COMP fragment (Gln505–Ala757), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Abundantly expressed in the chondrocyte extracellular matrix, and is also found in bone, tendon, ligament and synovium and blood vessels. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
