Product Overview
Clone AIC-3 is a rabbit monoclonal antibody that recognizes cytochrome c oxidase subunit 4 isoform 1, mitochondrial (gene COX4I1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 19.6 kDa, and the supplier reports an observed band at about 17 kDa.
COX4I1 is the widely expressed isoform of subunit 4 of cytochrome c oxidase (complex IV), the terminal enzyme of the mitochondrial respiratory chain that transfers electrons to oxygen. As a nuclear-encoded regulatory subunit it is commonly used as a mitochondrial loading control and marker in Western blot and imaging studies.

Samples with a confirmed band (WB): human HepG2 cells, human HeLa cells, human SW620 cells, human Caco-2 cells, rat heart tissue, mouse heart tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:1000 overnight at 4 °C, HRP-conjugated secondary antibody at 1:1000, ECL detection.
Samples with confirmed staining (IHC): human cervical cancer tissue, human liver cancer tissue, rat heart tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human COX IV.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places cytochrome c oxidase subunit 4 isoform 1, mitochondrial in the mitochondrion inner membrane. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
