Product Overview

This rabbit polyclonal antibody targets creatine kinase M-type (gene CKM) in human, mouse and rat samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 43.1 kDa, and the supplier reports an observed band at about 43 kDa.

Creatine kinase M-type (CKM) is the muscle isoform of creatine kinase, which reversibly transfers phosphate between ATP and creatine. It maintains ATP supply during muscle contraction, and the CK-MM and CK-MB enzymes are classic indicators of skeletal and cardiac muscle damage.

Validated Applications

Western blot 0.1–0.5 µg/mL
IHC (paraffin sections) 0.5–1 µg/mL
ELISA 0.1–0.5 µg/mL

Samples with a confirmed band (WB): human K562 cells, human CNE cells, human COLO-320 cells, rat heart tissue, rat skeletal muscle tissue, rat lung tissue, mouse heart tissue, mouse lung tissue, mouse skeletal muscle tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:10000, ECL detection.

Samples with confirmed staining (IHC): human heart tissue, mouse heart tissue, rat heart tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).

Immunogen

Recombinant human Creatine Kinase MM fragment (Phe3–Lys381), expressed in E. coli.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places creatine kinase M-type in the cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL; IHC (paraffin sections) 0.5–1 µg/mL; ELISA 0.1–0.5 µg/mL. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on goat samples?
A.Reactivity with goat samples has not been tested by the supplier. Aligning the immunogen sequence with the goat ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 43.1 kDa; observed band at about 43 kDa.
Q.What immunogen was used to raise this antibody?
A.Recombinant human Creatine Kinase MM fragment (Phe3–Lys381), expressed in E. coli.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
