Product Overview

Clone DCO-3 is a rabbit monoclonal antibody that recognizes cyclic AMP-responsive element-binding protein 1 (gene CREB1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 36.7 kDa, and the supplier reports an observed band at about 45 kDa.

CREB (CREB1) is a transcription factor that binds cAMP response elements (CRE) in gene promoters. Phosphorylation at Ser133 by PKA and other kinases recruits the coactivators CBP and p300 to switch on target genes, and CREB is widely studied in neuronal plasticity, memory, metabolism and cell survival.

Samples with a confirmed band (WB): NIH/3T3 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): mouse brain tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Samples with confirmed staining (ICC/IF): HeLa cells.

Immunogen

Synthetic peptide based on the sequence of human CREB.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places cyclic AMP-responsive element-binding protein 1 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.Has this antibody been tested on canine samples?
A.Reactivity with canine samples has not been tested by the supplier. Aligning the immunogen sequence with the canine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 36.7 kDa; observed band at about 45 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human CREB.
