Product Overview
This rabbit polyclonal antibody targets C-terminal-binding protein 2 (gene CTBP2) in human, mouse and rat samples and is validated for flow cytometry, ICC/IF, IHC and Western blot. The predicted molecular weight is 48.9 kDa, and the supplier reports an observed band at about 50 kDa.
CtBP2 is a transcriptional corepressor closely related to CtBP1 that binds diverse DNA-binding repressors and helps silence their target genes. An alternative isoform, RIBEYE, is a main structural protein of ribbon synapses in retinal photoreceptors and inner-ear hair cells, so CTBP2 antibodies are widely used as ribbon synapse markers.

Samples with a confirmed band (WB): human A549 cells, human PC-3 cells, human THP-1 cells, human HEK293 cells, human Caco-2 cells, human HeLa cells, rat C6 cells, mouse NIH/3T3 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human intestinal cancer tissue, rat skeletal muscle tissue, mouse skeletal muscle tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in citrate buffer (pH 6).
Samples with confirmed staining (ICC/IF): U2OS cells.
Samples with confirmed staining (ICC/IF): human colon cancer tissue, mouse intestine tissue.

Recombinant human CTBP2 fragment (His321–Gln445), expressed in E. coli. Sequence identity with the mouse and rat orthologs: 99.2% and 98.4%, respectively.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt places C-terminal-binding protein 2 in the nucleus and synapse. Tissue expression noted by UniProt (human): Ubiquitous. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
