Product Overview
Clone ADIA-3 is a rabbit monoclonal antibody that recognizes cullin-1 (gene CUL1) in human, mouse and rat samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 89.7 kDa, and the supplier reports an observed band at about 85 kDa.
Cullin-1 (CUL1) is the scaffold of SCF (SKP1-CUL1-F-box) E3 ubiquitin ligases. It binds SKP1 and an F-box protein at one end and the RING protein RBX1 at the other, bringing substrates close to the E2 enzyme. Activity depends on NEDD8 modification, and SCF ligases degrade many cell cycle and signaling proteins, making CUL1 central to protein turnover research.

Samples with a confirmed band (WB): human THP-1 cells, human K562 cells, human SIHA cells, human 293T cells, rat C6 cells, rat RH35 cells, mouse HEPA1–6 cells.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): human lung cancer tissue, human glioma tissue, human bladder cancer tissue, human breast cancer tissue, human colon cancer tissue, mouse brain tissue, rat brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human cullin 1.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. Tissue expression noted by UniProt (human): Expressed in lung fibroblasts. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
