Product Overview

Clone 19C43 is a rabbit monoclonal antibody that recognizes cullin-2 (gene CUL2) in human, mouse and rat samples and is validated for Western blot, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 87.0 kDa, and the supplier reports an observed band at about 87 kDa.

Cullin-2 (CUL2) is the scaffold of CRL2 E3 ubiquitin ligases, which use Elongin B/C and substrate adaptors carrying a VHL box. The best-known is the VHL-CUL2 ligase that targets HIF-α for degradation when oxygen is plentiful. Its activity requires neddylation, and CUL2 is studied in hypoxia signaling, protein turnover and targeted protein degradation.

Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:50
Flow cytometry 1:50

Samples with a confirmed band (WB): human Raji cells, human HepG2 cells, human PC-3 cells, rat skeletal muscle tissue, mouse skeletal muscle tissue.

Recommended loading (WB): 20–40 µg of total protein per lane.

Conditions in the example images (WB): 10% SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.

Immunogen

Synthetic peptide based on the sequence of human cullin 2.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places cullin-2 in the nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
