Biological background

CUL3 (protein: T-cell surface glycoprotein CD4) is a commonly studied target in molecular and cellular biology. Functional context (as provided): Essential for the control of the cell cycle at the G2/M (mitosis) transition. . Reported cellular localization context: Cytoplasm. Nucleus. Cytoplasm, cytoskeleton, microtubule organizing center, centrosome. Tissue expression notes (as provided): Ubiquitous. Expressed at high levels in testis.

Research relevance and current trends

  • Research context keywords from the source record include: Apoptosis,Cancer,Cell Biology,Cell Cycle,Cell Cycle Inhibitors,Cell Death,Nucleus,Proteasome / Ubiquitin,Proteolysis/Ubiquitin,SCF Complex E3 Ligase,Ubiquitin E3 Enzymes.
  • Current studies often focus on connecting target abundance/localization to pathway perturbations across models, tissues, and cell states.
  • Quantitative and multiplexed assays (e.g., imaging + immunoblot panels) are commonly used to compare phenotypes across conditions and time-courses.

Common research applications

  • Western blotting (WB): assess relative target abundance across samples, treatments, or time-points.
  • Flow cytometry: quantify target-positive populations and compare shifts in marker distributions.
  • Immunoprecipitation (IP): enrich target complexes for downstream immunoblot or interaction analyses.

Workflow ideas (metafield): Validate CUL3 antibody specificity using KO/KD control samples (WB/IF/IHC as appropriate), Detect CUL3 expression by Western blot in cell or tissue lysates, Quantify CUL3-positive cells by flow cytometry in single-cell suspensions, Enrich CUL3 by immunoprecipitation from lysates for downstream analysis

Notes for experimental interpretation

  • Consider isoforms and post-translational modifications (PTMs) that may shift apparent molecular weight or epitope accessibility.
  • Apparent molecular weight may vary by sample type and processing (observed MW: 133 kDa; calculated MW: 48337 MW).
  • Control concepts: include appropriate negative controls (e.g., isotype, KO/KD samples) and orthogonal validation when feasible.

Additional product details (from the source record)

  • Molecular weight (observed): 133 kDa
  • Cellular localization (provided): Cytoplasm. Nucleus. Cytoplasm, cytoskeleton, microtubule organizing center, centrosome.
  • Tissue details (provided): Ubiquitous. Expressed at high levels in testis.

Can you help my question with product M00747, anti-Cullin 3 Monoclonal antibody. I was wondering if it would be possible to conjugate this antibody with biotin. I would need it to be without BSA or sodium azide. I am planning on using a buffer exchange of sodium azide with PBS only. Would there be problems for me to conjugate the antibody and store it in -20 degrees in small aliquots?
We do not advise storing this antibody with PBS buffer only in -20 degrees. If you want to store it in -20 degrees it is best to add some cryoprotectant like glycerol. If you want carrier free M00747 anti-Cullin 3 Monoclonal antibody, we can provide it to you in a special formula with trehalose and/or glycerol. These molecules will not interfere with conjugation chemistry and provide a good level of protection for the antibody from degradation. Please be sure to specify this in your purchase order.
