Product Overview
This rabbit polyclonal antibody detects stromal cell-derived factor 1 (gene CXCL12) in mouse and rat samples and is validated for IHC and Western blot. The predicted molecular weight is 10.7 kDa, and the supplier reports an observed band at about 11 kDa.
CXCL12 (SDF-1) is a chemokine that binds the receptors CXCR4 and ACKR3. It retains hematopoietic stem cells in the bone marrow niche, guides lymphocyte and monocyte migration, and directs cell movement during embryonic development, including neural, vascular and germ cell migration. It is also studied in cancer metastasis and tissue repair.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 2–5 µg/mL |
Samples with a confirmed band (WB): rat stomach tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with confirmed staining (IHC): mouse spleen tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide from the C-terminal region of mouse CXCL12. Sequence identity with the human and rat orthologs: 96.2% and 95.5%, respectively.
Reactivity Notes
The supplier lists reactivity with mouse and rat. UniProt describes stromal cell-derived factor 1 as a secreted protein. Tissue expression noted by UniProt (human): Isoform Alpha and isoform Beta are ubiquitously expressed, with highest levels detected in liver, pancreas and spleen. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
