Product Overview

This rabbit polyclonal antibody detects C-X-C chemokine receptor type 4 (gene CXCR4) in human samples and is validated for Western blot. The predicted molecular weight is 39.7 kDa, and the supplier reports an observed band at about 50 kDa.

CXCR4 is a G protein-coupled receptor for the chemokine CXCL12 (SDF-1). It controls retention and homing of hematopoietic stem cells in the bone marrow and guides lymphocyte trafficking and embryonic cell migration. CXCR4 is also a coreceptor for T-tropic HIV-1 and is widely studied in cancer metastasis and stem cell mobilization.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human)

Samples with a confirmed band (WB): MDA-MB-231 cells, MCF-7 cells, Jurkat cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide from the N-terminal region of human CXCR4.

Reactivity Notes

The supplier lists reactivity with human. UniProt places C-X-C chemokine receptor type 4 in the cell membrane, cell junction, early endosome and late endosome. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on bovine samples?
A.Reactivity with bovine samples has not been tested by the supplier. Aligning the immunogen sequence with the bovine ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 39.7 kDa; observed band at about 50 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
