Key elements and design rationale

  • Antibody format: Rabbit Polyclonal Rabbit IgG
  • Immunogen / epitope context: E.coli-derived human Cyclin A1 recombinant protein (Position: D64-E263). Human Cyclin A1 shares 78.5% and 79.5% amino acid (aa) sequence identity with mouse and rat Cyclin A1, respectively. (reported region: D64-E263).
  • Molecular weight context: reported MW: 52 kDa; calculated MW: 52358 MW
  • Reactivity: Human,Mouse,Rat
  • Applications: Flow Cytometry, IF, ICC, WB

As a polyclonal antibody, the reagent recognizes multiple epitopes on the target, which can improve detection robustness but may increase sensitivity to sample-dependent epitope changes.

Biological background

Cyclin-A1; Cyclin-A1. Cyclin-A1 is a protein that in humans is encoded by the CCNA1 gene. The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance through the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. The cyclin encoded by this gene was shown to be expressed in testis and brain, as well as in several leukemic cell lines, and is thought to primarily function in the control of the germline meiotic cell cycle. This cyclin binds both CDK2 and CDC2 kinases, which give two distinct kinase activities, one appearing in S phase, the other in G2, and thus regulate separate functions in cell cycle. Also this cyclin was found to bind to important cell cycle regulators, such as Rb family proteins, transcription factor E2F-1, and the p21 family proteins. Multiple transcript variants encoding different isoforms have been found for this gene. Functional note: May be involved in the control of the cell cycle at the G1/S (start) and G2/M (mitosis) transitions. May primarily function in the control of the germline meiotic cell cycle and additionally in the control of mitotic cell cycle in some somatic cells. . Reported localization: Nucleus . Expression/tissue context: Very high levels in testis and very low levels in brain. Also found in myeloid leukemia cell lines.

Research relevance and current trends

  • Apoptosis: Researchers commonly examine how CCNA1 (Cyclin-A1) relates to this theme using model systems and orthogonal readouts.

Common research applications

  • Western blotting: compare relative CCNA1 (Cyclin-A1) levels across conditions; band patterns may reflect isoforms and processing.
  • IF/ICC: evaluate subcellular localization and co-localization patterns; signal can depend on fixation/permeabilization and epitope accessibility.
  • Flow cytometry: quantify target-positive populations and shifts in expression; gating strategy and background staining controls are essential.

Notes for experimental interpretation

  • Specificity notes: No cross reactivity with other proteins.
  • Cross-reactivity: No cross-reactivity with other proteins
  • Family / similarity context: Belongs to the cyclin family. Cyclin AB subfamily.
  • Isoforms and PTMs: Apparent size and signal patterns can differ across splice isoforms, proteolytic processing, and post-translational modifications.
  • Controls: Include an isotype control (as relevant), no-primary control for imaging, and orthogonal validation such as KD/KO samples when available.
