Product Overview

Clone CIC-3 is a rabbit monoclonal antibody that recognizes G2/mitotic-specific cyclin-B1 (gene CCNB1) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 48.3 kDa, and the supplier reports an observed band at about 60 kDa.

Cyclin B1 (CCNB1) binds and activates CDK1 to form maturation-promoting factor, the kinase that triggers entry into mitosis. Its levels rise through G2 and it is destroyed at the metaphase-to-anaphase transition, so it is a standard marker of G2/M cells in cell cycle and cancer studies.

Validated Applications

Western blot 1:1000–1:2000
Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:50
Flow cytometry 1:30

Samples with confirmed staining (IHC): human breast cancer tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human cyclin B1.

Reactivity Notes

The supplier lists reactivity with human. UniProt places G2/mitotic-specific cyclin-B1 in the cytoplasm and nucleus. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:1000–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:30; Immunoprecipitation 1:50. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 48.3 kDa; observed band at about 60 kDa. A difference from the calculated size is common and can reflect post-translational modification, processing or isoform differences.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide based on the sequence of human cyclin B1.
Q.How should this antibody be stored?
A.Long term: −20 °C for up to 12 months. Frequent use: 4 °C for up to 1 month. Avoid repeated freeze–thaw cycles.
