Product Overview

This rabbit polyclonal antibody detects cyclin-dependent kinase 7 (gene Cdk7) in human, mouse and rat samples and is validated for Western blot. The predicted molecular weight is 39.0 kDa, and the supplier reports an observed band at about 39 kDa.

CDK7 is the kinase at the core of the CDK-activating kinase (CAK), which it forms with cyclin H and MAT1. It activates other CDKs such as CDK1 and CDK2 by phosphorylating their T-loop, and as part of the general transcription factor TFIIH it phosphorylates RNA polymerase II, linking cell cycle control to transcription.

Validated Applications

Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse)

Samples with a confirmed band (WB): rat testis tissue, rat ovary tissue, HeLa cells, MCF-7 cells, A549 cells, COLO320 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Immunogen

Synthetic peptide from the C-terminal region of mouse cdk7. The peptide sequence is conserved in rat. It differs from the human sequence at two residues.

Reactivity Notes

The supplier lists reactivity with human, mouse and rat. UniProt places cyclin-dependent kinase 7 in the nucleus and cytoplasm. The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide and thimerosal (a mercury compound) as preservatives. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.

A.Validated applications with suggested starting dilutions: Western blot 0.1–0.5 µg/mL (Human, Rat, Mouse). Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human, Mouse, Rat. Other species are not listed by the supplier.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 39.0 kDa; observed band at about 39 kDa.
Q.What immunogen was used to raise this antibody?
A.Synthetic peptide from the C-terminal region of mouse cdk7. The peptide sequence is conserved in rat.
Q.How should this antibody be stored?
A.As supplied: −20 °C for up to 12 months from receipt. After reconstitution: 4 °C for up to 1 month, or aliquot and keep at −20 °C for up to 6 months. Avoid repeated freeze–thaw cycles.
