Product Overview
Clone AABG-16 is a rabbit monoclonal antibody that recognizes peptidyl-prolyl cis-trans isomerase A (gene PPIA) in human, mouse and rat samples and is validated for Western blot and IHC. The predicted molecular weight is 18.0 kDa, and the supplier reports an observed band at about 18 kDa.
Cyclophilin A (PPIA) is an abundant peptidyl-prolyl cis-trans isomerase that helps proteins fold by switching the configuration of proline bonds. It is the intracellular target of cyclosporin A. When secreted, cyclophilin A acts on its receptor CD147 to attract leukocytes and activate inflammatory signaling, and it is often used as a loading control.

Validated Applications
| Western blot | 1:500–1:2000 |
|---|---|
| Immunohistochemistry | 1:50–1:200 |
Samples with a confirmed band (WB): human HepG2 cells, human HUVEC cells, human HeLa cells, human THP-1 cells, rat kidney tissue, rat brain tissue, mouse kidney tissue, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 1:500 overnight at 4 °C, HRP-conjugated secondary antibody at 1:500, ECL detection.
Samples with confirmed staining (IHC): human colon cancer tissue, human stomach cancer tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Synthetic peptide based on the sequence of human cyclophilin A.
Reactivity Notes
The supplier lists reactivity with human, mouse and rat. UniProt describes peptidyl-prolyl cis-trans isomerase A as secreted and also located in the cytoplasm, nucleus and cell membrane. The samples tested by the supplier (listed above) are a practical starting point for positive controls.
