Product Overview
This rabbit polyclonal antibody recognizes cystatin-C (gene CST3) in human and mouse samples and is validated for ELISA, IHC and Western blot. The predicted molecular weight is 15.8 kDa, and the supplier reports an observed band at about 16 kDa.
Cystatin C (CST3) is a small secreted inhibitor of cysteine proteases, including cathepsins, made by nearly all nucleated cells. Because it is produced at a steady rate and cleared by glomerular filtration, its level in blood is widely used to estimate kidney function, and it is also studied in vascular and neurodegenerative disease.

Validated Applications
| Western blot | 0.1–0.5 µg/mL |
|---|---|
| IHC (paraffin sections) | 0.5–1 µg/mL |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human COLO-320 cells, human placenta tissue, human HepG2 cells, mouse brain tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 50 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL, HRP-conjugated secondary antibody at 1:10000, ECL detection.
Samples with confirmed staining (IHC): mouse brain tissue.
Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.
Conditions in the example images (IHC): heat-mediated antigen retrieval in EDTA buffer (pH 8.0).
Immunogen
Recombinant human Cystatin C fragment (Lys31–Ala146), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt describes cystatin-C as a secreted protein. Tissue expression noted by UniProt (human): Expressed in submandibular and sublingual saliva but not in parotid saliva (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
