Product Overview

Clone EEO-11 is a rabbit monoclonal antibody that recognizes keratin, type I cytoskeletal 14 (gene KRT14) in human samples and is validated for Western blot, IHC, ICC/IF, immunoprecipitation and flow cytometry. The predicted molecular weight is 51.6 kDa, and the supplier reports an observed band at about 47 kDa.

Keratin 14 (KRT14) is a type I intermediate filament protein of the basal, proliferating layer of stratified epithelia. It pairs with keratin 5 to form bundled filaments that anchor to hemidesmosomes and desmosomes and resist mechanical stress. KRT14 is a common basal cell marker, and its mutations cause epidermolysis bullosa simplex, a skin blistering disorder.

Immunohistochemistry 1:50–1:200
Immunocytochemistry / Immunofluorescence 1:50–1:200
Immunoprecipitation 1:500
Flow cytometry 1:50

Samples with a confirmed band (WB): A431 cells.

Recommended loading (WB): 20–40 µg of total protein per lane.

Samples with confirmed staining (IHC): human skin tissue.

Recommended antigen retrieval: heat-mediated, in TE buffer (pH 9.0); citrate buffer (pH 6.0) can be used instead.

Immunogen

Synthetic peptide based on the sequence of human cytokeratin 14.

Reactivity Notes

The supplier lists reactivity with human. UniProt places keratin, type I cytoskeletal 14 in the cytoplasm and nucleus. Tissue expression noted by UniProt (human): Expressed in the corneal epithelium (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.

Safety

Handling: the formulation contains sodium azide as a preservative. Wear gloves, avoid contact with skin and eyes, and dispose of waste according to local regulations. For research use only; not for diagnostic or therapeutic procedures.
Q.Which applications is this antibody validated for, and at what dilution?
A.Validated applications with suggested starting dilutions: Western blot 1:500–1:2000; Immunohistochemistry 1:50–1:200; Immunocytochemistry / Immunofluorescence 1:50–1:200; Flow cytometry 1:50; Immunoprecipitation 1:500. Optimal dilutions should be determined by each laboratory.
Q.Which species does this antibody react with?
A.Validated reactivity: Human. Other species are not listed by the supplier.
Q.Has this antibody been tested on horse samples?
A.Reactivity with horse samples has not been tested by the supplier. Aligning the immunogen sequence with the horse ortholog (e.g., by BLAST) helps estimate the chance of cross-reactivity.
Q.Is a blocking reagent available for this antibody?
A.A matching blocking reagent for competition (specificity) controls can be supplied on request. Contact info@biohippo.com to request it.
Q.Is a carrier-free (BSA-free) version available, for example for conjugation?
A.Yes. A carrier-free formulation without BSA, stabilized with trehalose and/or glycerol, can be prepared on request and is compatible with common conjugation chemistries. Please note this when ordering or contact info@biohippo.com. For storage at −20 °C, keep a cryoprotectant such as glycerol in the buffer rather than PBS alone.
Q.What band size should be expected in Western blot?
A.Reported by the supplier: calculated molecular weight 51.6 kDa; observed band at about 47 kDa.
