Product Overview
This rabbit polyclonal antibody targets D-amino-acid oxidase (gene DAO) in human and mouse samples and is validated for Western blot, flow cytometry and ELISA. The predicted molecular weight is 39.5 kDa, and the supplier reports an observed band at about 39 kDa.
D-amino acid oxidase (DAO) is a peroxisomal flavoenzyme that breaks down D-amino acids by oxidative deamination, producing keto acids, ammonia and hydrogen peroxide. In the brain it degrades D-serine, a co-agonist of NMDA receptors, so it can influence glutamate signaling. It is studied in neuroscience, schizophrenia research and in kidney handling of dietary D-amino acids.

Validated Applications
| Western blot | 0.25–0.5 µg/mL (Human, Mouse) |
|---|---|
| Flow cytometry (fixed cells) | 1–3 µg/1×106 cells (Human) |
| ELISA | 0.1–0.5 µg/mL |
Samples with a confirmed band (WB): human hcct tissue, mouse kidney tissue.
Recommended loading (WB): 20–40 µg of total protein per lane.
Conditions in the example images (WB): 5–20% gradient SDS-PAGE under reducing conditions, 30 µg lysate per lane, transfer to nitrocellulose membrane, blocking in 5% non-fat milk, primary antibody at 0.5 µg/mL overnight at 4 °C, HRP-conjugated secondary antibody at 1:5000, ECL detection.
Samples with a confirmed signal (flow cytometry): U937 cells.
Immunogen
Recombinant human DAO fragment (Cys18–Lys338), expressed in E. coli.
Reactivity Notes
The supplier lists reactivity with human and mouse. UniProt describes D-amino-acid oxidase as secreted and also located in the peroxisome matrix, cytoplasm and presynaptic active zone. Tissue expression noted by UniProt (human): Expressed in the cerebellum, in astrocytes of the cortex, in motor neurons and fibers of the lumbar spinal cord (at protein level). The samples tested by the supplier (listed above) are a practical starting point for positive controls.
